چکیده
Dutch iris is a commercially important bulbous ornamental crop. Its high demand in global floriculture market necessitates the production of its high-quality planting material. In present investigation, an efficient vitro propagation system has been developed for Iris × hollandica Tub. cv. Professor Blaauw (Dutch iris) using meta-Topolin (mT) first time. Effect various concentrations BAP, Kn, and mT (0, 0.5, 1.0, 1.5 2 mg L−1) along with varying photoperiods (16 h light dark incubation 1, 2, 3 4 weeks) on shoot induction from twin scale explants was studied. Of cytokinins tested, different doses resulted better response than BAP Kn. Photoperiod duration also affected significantly. Along 1 week, cytokinin at 1.0 L−1 MS medium maximum (91.63%) increased emergence micro shoots (4.83 shoots/explant average length 5.02 cm). Efficacy alone or combination auxins multiplication compared. The synergistic effect cytokinin-auxin comprising + 0.25 NAA considerably higher number (17.53) mean (7.06 cm) bulblets (2.74). Positive sucrose concentration (90 g paclobutrazol (5 bulblet formation size observed respectively. superiority over found during rhizogenesis. Shoots raised were healthy long enough, thus showed rooting (63.83%) ½ 0.5 IAA after weeks incubation. About 89.16% survival rate recorded plantlets under ex conditions. Analysis clonal fidelity thirteen regenerated plants done SCoT markers. Out 36 primers, 13 primers clearly scorable monomorphic bands, displaying genetic uniformity among plantlets. This mediated protocol can be routinely used rapid large this valuable Reliable consistent regeneration established, which applied crop will open up way engineering/gene editing other crops.

Type-1 ribosome-inactivating protein from iris (Iris hollandica var. Professor Blaauw) binds specific genomic DNA fragments.
The capacity of IRIP, a type-1 ribosome-inactivating protein (RIP) isolated from the bulbs of Iris hollandica, to bind specific DNA sequences from a mixture of approx. 200 bp (average length) fragments of total genomic DNA from Iris genome was studied. Fragments that were preferentially bound by IRIP were enriched by several cycles of affinity binding and PCR, and were cloned and sequenced. The...
متن کاملType 1 ribosome-inactivating proteins are the most abundant proteins in iris (Iris hollandica var. Professor Blaauw) bulbs: characterization and molecular cloning.
The most abundant protein of Iris bulbs has been identified as a type 1 ribosome-inactivating protein (RIP). Analysis of the purified proteins and molecular cloning of the corresponding cDNAs demonstrated that this type 1 RIP is a mixture of three isoforms that exhibit a high degree of sequence identity and have similar, though not identical, ribosome-inactivating and polynucleotide:adenosine g...

Callus Induction and Plant Regeneration of Chrysanthemum morifolium and C. coccineum via Direct and Indirect Organogenesis and Genetic Fidelity Analysis Using IRAP, ISSR and SCoT Molecular Markers
In vitro propagation of C. morifolium cv. ‘Homa’ and cv. ‘Delkash’ and wild C. coccineum via direct and indirect organogenesis and somatic embryogenesis were investigated. BAP at 0, 1, 2 and 3 mg l-1 or NAA at 0, 0.05, 0.1 and 0.2 mg l-1 concentrations were used to induce direct and indirect organogenesis of shoot tip explants. To study the callu...
